Abstract

Characterisation and identification of proteins and peptides using amino acid analysis, N-terminal sequencing, high performance liquid chromatography (HPLC) and mass spectrometry (MS).Overview on protein and proteom analytics and focussing on modern methods.

Objective

- Handling of very small amounts (< picomoles) of protein and peptide material- Understand the basic principles of mass spectrometry - Protein and peptide specific sample preparation for MALDI and ESI MS - Basic operations of various analytical apparatus - Learn to apply programs developed for MS; e.g. Biotools

Potential types of experiments Experimental part 1: Characterisation of proteins: - Reductive alkylation of proteins - Disulfidemapping of proteins - Detect partially reduced protein specimen by HPLC and MS - Localize and identify artificially or naturally modified amino acids (chemical labels or post-translational modifications; e.g. phosphorylation sites) Analytical methods - Chemical and enzymatic fragmentation - Amino acid analysis - Edman sequencing - Capillary HPLC and MS coupling (LC-MALDI MS and LC-ESI MS) - Electrospray Ionisation (ESI) and Matrix Assisted Laser Desorption Ionisation (MALDI) MS - Tandem mass spectrometry (MS/MS)Experimental part 2: Identification of proteins from solution and after SDS PAGE: - In-gel digestion - Peptide mass fingerprinting (PMF) Analytical methods - Electrospray Ionisationa (ESI) and Matrix Assisted Laser Desorption Ionisation (MALDI) MS - Tandem MS (MS/MS) - Capillary HPLC and MS coupling (LC-MALDI MS and LC-ESI MS) - Protein identification by using Mascot and other search engines
