Abstract
Basics of light microscopy: preparation of plant material, best use of the microscope and documentation.The plant body: organs, tissues and cells. Anatomy and function of roots, stems and leaves.Adaption of plants to extreme habitats. Characteristics of the plant cell. Angiosperm reproduction. Seed and fruit development. Seed germination.
Objective
Skill in preparing botanical objects for microscopic studies. Getting the best image of which the microscope is capable. Understanding of the correlation between structure and function at the level of organs, tissues and cells. Bridging to plant systematics, physiology and ecology. Taking pleasure in the external and internal architecture of plants.
Content
Basics of light microscopy: eye, image formation by a convex lens, simple and compound microscopes. Marking of objectives, eyepieces and condensers. Köhler illumination. Image formation according to Abbe. Optical contrast methods (bright-field with axial and oblique illumination, dark-field, phase-contrast and polarization-contrast). Measuring methods. The plant body as a hierarchy of organs, tissues and cells. Anatomy and function of the three basic plant organs roots, stems and leaves. Staining procedures in plant histology and cytology. Adaption of plants to extreme habitats: xerophytes, hydrophytes and insectivores. Characteristics of the plant cell: plastids, cell wall and vacuole. Mitosis. Angiosperm reproduction: flower structure, gametophyte development, pollination and double fertilization, seed and fruit development, seed germination and storage material of endosperms and cotyledons.
