Abstract

Introduction to the theory and fundamentals of light microscopy. Students learn how to handle the light microscope safely and competently, as well as how to prepare microscopic specimens. They examine plant structures and document their observations systematically.

Objective

Upon completion of the course, students will be able to: - independently prepare biological specimens, operate light microscopes, and document microscopic observations clearly and systematically; - explain the relationship between plant structure and function at the cellular, tissue, and organ levels, and illustrate these correlations using concrete examples; - describe how plant anatomy is interconnected with systematics, physiology, ecology, and development, and discuss these links in a biological context.

Content

The course introduces the fundamentals of light microscopy and its application in plant anatomy. Topics include optical principles, the structure and function of the light microscope, Köhler illumination, as well as contrast-enhancing techniques. Students learn to prepare microscopic specimens and to document and measure plant structures accurately. The biological component focuses on the structure and function of plant cells, tissues, and selected organs — from the cell to stem and leaf. Particular emphasis is placed on plastids, vacuole, and cell wall, as well as on the anatomy of various plant tissues (e.g., epidermis, vascular tissues, wood). Anatomical adaptations to different environments are discussed using selected examples.
