Content

Block 1: Spectroscopical methods for the study of biological macromolecules [nuclear magnetic resonance (NMR), Electron Spin Resonance (ESR), fluorescence, and circular dichroism (CD) spectroscopy]. Block 2: Enzymatic and chemical fragmentation of proteins, SDS-polyacrylamide gel electrophoresis, electroblotting, amino-terminal sequencing of an unknown peptide using manual Edman degradation, high pressure liquid chromatography (HPLC), sequence analysis with the aid of data-bases, UV spectroscopy of proteins, quantitative determination of disulfide bonds, determination of the activation energy of non-catalyzed and and catalyzed reactions using starch degradation as an example. Block 3: Work on two small research projects. Block 4: Protein-DNA interactions and X-ray cristallography. Plasmid isolation, purification of DNA fragments, Analysis of the secondary structure of DNA and of transcription factors with DNA (gel retardation assay). overexpression of proteins in E.coli and enrichment via anion-exchanger, refolding of a multimeric protein complex from denatured components, preparation and analysis of a protein-DNA complex. X-ray crystallography of macromolecules, an overview: instrumentation, data analysis, computer-graphics.
